Objective To understand the distribution and genotype of Wolbachia in mosquito population in guangzhou. Methods Using 16 S r DNA and WSP gene sequence of Wolbachia, a PCR detection method was established to detect some species of mosquitoes of the Aedes, Culex, Armigeres and Toxorhynchites in the Culicidae Mosquitoes. Meanwhile, Mega6.06 software was used for phylogenetic analysis of the obtained gene sequence. Results A total of 4 common mosquitoes of 3 different habitats in Guangzhou were detected, and Wolbachia had a total infection rate of 98.75%, The detection rate of 16 S r DNA gene was highest in Aedes albopictus from the habitat of residential area(100%) and Culex quinquefasciatus wearied from the habitat of park(100%), followed by Armigeres obturbans from the habitat of park(94.44%); Aedes albopictus from the habitat of school had the lowest wsp gene detection rate, which was 90%. The nucleic acid positive rate of four mosquito species Wolbachia was 50% ~ 100%. The 16 S rDNA gene sequence of 14 entries was obtained with the sequence length of 266-404 bp. The sequence length of the 26-item wsp gene is 331-474 bp, and the phylogenetic tree shows that these sequences belong to two supergroups, A and B, respectively. The Wolbachia genotype is wAlbA, wAlbB or wAlbA and wAlbB. Conclusion Wolbachia is commonly infected by aedes albopictus, culex pipiens and aedes aegypti in Guangzhou. Its genotypes were wAlbAor wAlbB, or wAlbA and wAlbB. The genes of wolbachia may differ between different species of mosquito from different habitats.
