地球是大约1012种微生物的家园,但其中大部分微生物尚未被发现鉴定,不断发现鉴定和研究微生物新菌种是丰富微生物种质资源库和支撑微生物科技创新的重要基础。地球70%的表面为水生环境所覆盖,多样的水生环境中生存有大量未被发现的微生物类群。本论文研究了我国东海及高原盐湖两大水生环境中的细菌多样性,从中分离培养和鉴定了四百余株细菌菌株,以16S rRNA基因序列的系统发育分析为基础研究了东海和高原盐湖的微生物多样性,结果表明分离自东海海域的302株菌株中γ-变形菌(Gammaproteobacteria)细菌(64%)为最主要类群,其次是拟杆菌(Bacteroidetes)(11%),a-变形菌(Alphaproteobacteria)(9%),厚壁菌(Firmicutes)(8%),β-变形菌(Betaproteobacteria)(5%)和放线菌(Actinobacteria)(3%);而分离自高原盐湖的107株菌株中γ-变形菌(Gammaproteobacteria)细菌(29%)、a-变形菌(Alphaproteobacteria)(27%)和厚壁菌(Firmicutes)(22%)为主要类群,两类水生环境中的微生物多样性具有显著的差异。在分离的四百余株细菌中,其中58株细菌与已知菌株的16S rRNA基因序列相似度≤98.65%,为潜在细菌新种。本研究对其中三株黄杆菌属潜在新种进行了多项类系统鉴定,分别鉴定命名为Flavobacterium sharifuzzamanii,F.zhairuonensis 和 F.chakense,同时应用全基因组测序、基因簇预测、微生物发酵及抗菌活性实验等对F.zhairuonensis产生次级代谢产物潜能进行了初步的分析研究。本研究为我国东海及高原盐湖的微生物多样性研究提供了参考,同时分离鉴定的细菌新菌株丰富了微生物种质资源,为后续开发应用海洋环境及盐湖微生物资源奠定基础。
