Peronophythora litchii causes downy blight on litchi fruits as well as tender leaves and panicles rot of litchi plants. RXLR effectors secreted by Phytophthora species play a central role in pathogen–plant interactions.Though 245 RXLR effectors have already been predicted in this litchi downy blight pathogen before, the function of them is still unknown. Here, we identified a RXLR effector PlAvh142 that could trigger strong cell death in Nicotiana benthamiana. In addition, heterologous expression experiments showed that PlAvh142 could induce ROS accumulation, callose deposition and genes transcription involved in plant hormone pathways in N.benthamiana. Next, We found that PlAvh142 localize both in cytoplasm and nucleus. The cytoplasmic localization was required for its cell death-inducing activity. Moreover, deletion of the two internal repeats in PlAvh142 also abolished the cell death-inducing activity. Through aligning the ORF of thirty strains, five SNPs were found in PlAvh142 and only three of them led to nonsynonymous substitutions, but none of the version affect cell death. The results of expression pattern revealed that PlAvh142 is highly transcribed during zoospores and the early stages of infection. Finally in the pathogenicity assay, PlAvh142 knockout mutants attenuated P.litchii virulence on litchi plants, whereas the overexpressed mutants are more aggressive. In conclusion,PlAvh142 is an important virulence RXLR effector of P. litchii and recognized by N benthamiana.
