The VfmIH two-component system modulates multiple virulence traits in Dickeya zeae

Pathogenic bacteria Dickey a zeae strain EC1 produces a family of antibiotics-like phytotoxins named zeamines,which are encoded by the zms gene cluster and critical for bacterial virulence.In this study,we identified a zeamine deficient mutant with Tn5 inserted in a gene encoding a two-component system(TCS) sensor histidine kinase(HK).Bioinformatics analysis showed that this kinase gene is the homologue of the vfml gene of D.dadantii strain 3 937.Similar to the case of D.dadantii 3 937,a homologue of vfmH encoding a response regulator(RR) is located at the vicinity of vfml and sharing the same operon.Deletion in—frame of either vfmI or vfmH resulted in significantly reduced production of zeamines and cell wall degrading enzymes(CWDEs),and mitigated the bacterial virulence on rice seeds and potato tubers.RNA-seq and Q-RT PCR analysis of D.zeae EC1 and the mutant of VfmH showed that the TCS positively modulated the transcriptional expression of a range of virulence genes,including zms,T1 SS,T2 SS,T6 SS,flagellar and CWDEs genes.In D.dadantii 3937,VfmH protein was shown to interact with the promoters of vfmA and vfmE to affect the transcription.This study found that the VfmH protein not merely could directly interact with the promoters of vfmA and vfmE but also interact with the promoters of vfmF to affect the transport of Vfm signals.
