Most reported members of totiviridae lack an extracellular life cycle stage and are transmitted by cytoplasmic interchange. However, several novel unassigned viruses related to Totiviridae were reported in recent years to exhibit the extracellular transmission pathway, including the previously reported Omono River Virus（OmRV） with no protrusions separated from mosquito.Here a new OmRV strain assigned OmRV-LZ was identified and structurally determined by Cryoelectron microscopy reconstruction. Two 3-dimensional（3-D） maps with or without fiber-like protrusions were obtained by 3-D classification. Our results demonstrated the protrusion that has a weak connection with capsid protein subunit A and may be easily broken. It is suggested that the protrusion is formed by the ORF1-encoded small protein, and may play roles in cell entry.
