Screening of effector candidates from Sclerotinia sclerotiorum

Sclerotinia sclerotiorum(Lib.) de Bary is a necrotrophic plant pathogenetic fungus with broad host range.It also causes yield losses and quality decline in crop production.Studies on pathogenesis mechanism of S.sclerotiorum can provide potential targetfor Sclerotinia disease management and offer a reference basis for breeding of resistant plant.The interaction of necrotrophic with theirhosts is more complex than initially thought,and still poorly understood.We combined bioinformatics approaches and transcriptome analysis to determine the repertoire of S.sclerotiorum effector candidates and conducted detailed sequence and expression analyses on selected candidates.We identified 34 S.sclerotiorum secreted protein genes(SsEC1-34) expressed in planta,many of which have no predicted enzymatic activity,was up-regulated during the infection period and may be involved in the interaction between the fungus and its hosts.To elucidate the localization and possible functions of SsECs in plant cells,we conducted heterologous expression analysis in N.benthamiana through agroinfiltration method including full length(FL) or without signal peptide(NS) fusion with green fluorescent protein(GFP).The fluorescence signal of 4 effectors(FL and NS),including SsEC10,27,29 and 34 were detected in nucleusand cytoplasm,SsEC25,26,28 were also accumulated in cytoplasm.SsEC24(FL) were expressed inapoplast.Additionally,in 3 immune marker genes,the transcript levels of NbPRl,NbPR2,and NbPR4 were down-regulated by SsEC34 expression,whereasthe transcript levels of NbPRl and NbPR4 were up-regulated by SsEC24 expression.Interestingly,SsEC24(FL) could inducenecrosis like cell death,bioinformatics analysis showed that it is anelicitor-like protein.We speculate that it may act as a Pathogen-associated molecular patterns(PAMPs) activated host immune response.
