Development of rapid detection methods for African Swine Fever virus

The first case of African swine fever(ASF) was detected in Shenyang last August in China. Until April of this year, more than 120 outbreaks across 30 provinces, autonomous regions and municipalities were reported by the government. With no effective vaccines and treatment, rapid and accurate diagnostic methods are critical to the control of the rapid spreading disease. In this study, we developed three molecular biological techniques, real-time PCR(qPCR), nanoparticles PCR(Nano-PCR) and loop-mediated isothermal amplification(LAMP) combined with TaqMan probe(LAMPTaqMan) targeting the conserved gene, p72 or p54, of the ASF virus. The sensitivity and specificity of the three methods were analyzed. Our results showed that the detection limitation of qPCR, Nano-PCR and LAMP-TaqMan were 10 copies/μl, 10 copies/μl and 5 copies/μl of the standard plasmid, respectively, after optimization of the reaction conditions. The three assays showed no cross-reactivity with other swine virus. Moreover, the LAMP-TaqMan assay could be done in less than half an hour. Either DNA, blood or tissue samples could be detected by the LAMP-TaqMan assay with or without the simple pretreatment by specific solutions. Our results demonstrated that qPCR, Nao-PCR and LAMP-TaqMan are effective methods for rapid and accurate detection of ASFV which with great potential for developing diagnostic approaches.
