A convenient gene deletion toolbox for genetic analysis in Ralstonia solanacearum

Ralstonia solanacearum,a soilborne bacterial pathogen,is the causal agent of bacterial wilt and infects over 200 plant species in 50 families.The pathogen is lethal to many solanaceous species,including eggplant,tobacco and peanut.The pathogen is notorious for its genetic variations,and the regulatory mechanism that governs the pathogenicity of R.solanacearum is not well—understood.In this study,we set to develop a convenient and reliable delivery method for stable and directed deletion of gene or DNA sequence in the genome of R.solanacearum.The method is modified from the suicide vector pRC,which was developed previously for gene knock—in at a permissive site downstream of glmS in the genome of R.solanacearum strain GMI1000 through homologous recombination.The modified gene deletion system consists of a fusion DNA fragment containing the upstream and downstream region of a target gene and a gentamycin resistance gene as a selection marker,as well as a growth medium which primes R.solanacearum to a state prone to receiving DNA fragments for integration.The feasibility of this gene deletion system was tested by targeting a regulatory gene in R.solanacearum EP1,which is a virulent strain isolated from the diseased eggplant in Guangdong Province of China.The results showed that the resultant mutants displayed much reduced EPS production.Development of such a convenient gene deletion system would facilitate further investigations of the pathogenic mechanisms of this important bacterial pathogen.
