论文摘要
【目的】为研究辣椒耐盐品种,提高耐盐性和抗旱性,对干制辣椒(Capsicum annuum L.)CaNAC61基因进行克隆、生物信息学和表达水平分析。【方法】采用RT-PCR的方法,从辣椒中克隆得到CaNAC61基因,并分析该基因编码蛋白的理化性质、蛋白质二级和三级结构、系统进化树。采用qRT-PCR分析CaNAC61基因在胁迫处理下的表达情况。【结果】获得干制辣椒NAC转录因子CaNAC61(Genebank登录号:XM016719693.1)基因,该基因的开放阅读框长度为885 bp,编码294个氨基酸,蛋白质相对分子质量为33.67 kD,理论等电点为6.26。CaNAC61与不同植物中NAC家族成员同源性比对和进化树分析表明,CaNAC61属于NAC转录因子家族ATAF亚家族成员。荧光定量分析表明,CaNAC61在6种非生物胁迫处理下均有响应。其中,NaCl处理下表达上调较为显著,其次是干旱处理。【结论】本文成功克隆出CaNAC61基因,为研究辣椒NAC转录因子的功能提供参考依据。
